Microenvironment and Immunology TGFb Is a Master Regulator of Radiation Therapy- Induced Antitumor Immunity

چکیده
T cells directed to endogenous tumor antigens are powerful mediators of tumor regression. Recent immunotherapy advances have identified effective interventions to unleash tumor-specific T-cell activity in patients who naturally develop them. Eliciting T-cell responses to a patient's individual tumor remains a major challenge. Radiation therapy can induce immune responses to model antigens expressed by tumors, but it remains unclear whether it can effectively prime T cells specific for endogenous antigens expressed by poorly immunogenic tumors. We hypothesized that TGFb activity is a major obstacle hindering the ability of radiation to generate an in situ tumor vaccine. Here, we show that antibody-mediated TGFb neutralization during radiation therapy effectively generates CD8þ T-cell responses to multiple endogenous tumor antigens in poorly immunogenic mouse carcinomas. Generated T cells were effective at causing regression of irradiated tumors and nonirradiated lung metastases or synchronous tumors (abscopal effect). Gene signatures associated with IFNg and immune-mediated rejection were detected in tumors treated with radiation therapy and TGFb blockade in combination but not as single agents. Upregulation of programmed death (PD) ligand-1 and -2 in neoplastic and myeloid cells and PD-1 on intratumoral T cells limited tumor rejection, resulting in rapid recurrence. Addition of anti–PD-1 antibodies extended survival achieved with radiation and TGFb blockade. Thus, TGFb is a fundamental regulator of radiation therapy's ability to generate an in situ tumor vaccine. The combination of local radiation therapy with TGFb neutralization offers a novel individualized strategy for vaccinating patients against their tumors. Cancer Res; 75(11); 2232–42. 2015 AACR.

Gold nanoparticle-induced sonosensitization enhances the antitumor activity of ultrasound in colon tumor-bearing mice
Introduction: Light-driven cancer therapy strategies (e.g. photodynamic therapy and photothermal therapy) have undergone remarkable progress in recent years, but they still suffer from a serious drawback of limited penetration depth of light in tissue. As a non-invasive and non- ionizing radiation, ultrasound can be focused remotely, transferring acoustic energy deep in the bo...
متن کاملIL4 limits the efficacy of tumor-targeted antibody therapy in a murine model.
Tumor-targeted antibody therapy has had a major impact on reducing morbidity and mortality in a wide range of cancers. Antibodies mediate their antitumor activity in part by activating immune effector cells; however, the tumor microenvironment (TME) is enriched with cellular and soluble mediators that actively suppress generation of antitumor immunity. Here, we investigate the potential of pros...
متن کاملMicroenvironment and Immunology ATM-Mediated DNA Damage Signals Mediate Immune Escape through Integrin-avb3–Dependent Mechanisms
Although the tumor microenvironment plays a critical role in tumor progression and metastasis, the relationship between chemotherapy resistance andmodulation of the tumormicroenvironment remains unclear. Here, we report a novel mechanism showing how constitutive DNA damage signals in therapy-resistant tumor cells suppress antitumor immunity in an integrin-avb3–dependent manner. Integrin-avb3 wa...
متن کاملInhibition of Transforming Growth Factor-B1 Signaling Attenuates Ataxia Telangiectasia Mutated Activity in Response to Genotoxic Stress
Ionizing radiation causes DNA damage that elicits a cellular program of damage control coordinated by the kinase activity of ataxia telangiectasia mutated protein (ATM). Transforming growth factor B (TGFB)-1, which is activated by radiation, is a potent and pleiotropic mediator of physiologic and pathologic processes. Here we show that TGFB inhibition impedes the canonical cellular DNA damage s...
